Gut Health & Immunity Peptide Therapy โ€” Leaky Gut, IBS, IBD & Immune Support | InformedPeptides

Gut Health & Immunity

70% of your immune cells live in and around your gut lining. When that lining breaks down, the damage doesn't stay in your digestive system โ€” it reaches your brain, your skin, your joints, and your immune response. BPC-157 was literally discovered in the stomach. Restoring the gut is restoring your entire biological foundation.

Gut Health Immune System Gut-Brain Axis IBS & IBD
70%
Of the body's immune cells are housed in the gut-associated lymphoid tissue โ€” making the gut the largest immune organ
90%
Of the body's serotonin is produced in the gut โ€” making gut barrier integrity a direct determinant of mood
10โ€“15%
Of the global population meets criteria for IBS โ€” the most common functional gut disorder, and the one most underserved by conventional medicine
The Core Framework

The Gut Is Not Just a Digestive Organ โ€” It's Your Largest Immune Organ

Understanding this single fact reframes gut health from a digestive complaint into a whole-body priority.

The gastrointestinal tract houses approximately 70% of the body's total immune cells in a specialized tissue called the gut-associated lymphoid tissue (GALT). It's also the primary site of serotonin production, a key regulator of the autonomic nervous system, and home to trillions of microbes that collectively influence everything from inflammation to mood to metabolic health.

The entire gut is separated from everything else by a single layer of epithelial cells connected by proteins called tight junctions. When these tight junctions are intact, nutrients pass through while pathogens and bacterial fragments stay out. When they break down โ€” intestinal hyperpermeability, or "leaky gut" โ€” those unwanted substances enter the bloodstream directly. The 70% of immune cells living right there at the barrier respond immediately. Inflammation fires. And because that response is systemic, the consequences appear throughout the body โ€” often far from the gut itself.

BPC-157 Was Born in the Gut BPC-157 wasn't designed for gut healing โ€” it was discovered there. Isolated from human gastric juice at the University of Zagreb in 1991, it's a 15-amino acid peptide that the stomach produces naturally. It was evolved by the body to survive the gut environment and repair it from within. That's not a metaphor โ€” it's the literal biology of where this compound comes from. No other peptide in this stack has that pedigree.

The gut communicates bidirectionally with both the brain (vagus nerve) and the immune system (GALT). When the gut barrier fails, all three axes are disrupted simultaneously โ€” which is why gut healing produces benefits far beyond digestion.

The Ripple Effect

What Leaky Gut Looks Like Beyond the Gut

When tight junctions break down, the damage expresses throughout the body โ€” often in places that seem completely unrelated to digestion.

๐Ÿง
Brain Fog & Anxiety
Systemic inflammation from leaky gut crosses the blood-brain barrier. 90% of serotonin is gut-derived โ€” compromised lining means compromised mood and cognitive signaling from the source.
๐Ÿฆด
Joint Pain & Inflammation
LPS (lipopolysaccharide) fragments from gut bacteria entering the bloodstream trigger systemic inflammatory responses that localize in joints. Many "inflammatory arthritis" presentations have gut permeability as a primary upstream driver.
๐Ÿ”ด
Skin Conditions
Eczema, psoriasis, rosacea, and acne all have documented associations with gut permeability โ€” the gut-skin axis mirrors the gut-brain axis. Systemic inflammation from the gut expresses through the skin's immune layer.
๐Ÿ›ก๏ธ
Autoimmune Conditions
Molecular mimicry โ€” where the immune system mistakes body tissue for foreign proteins that look similar to gut-leaked particles โ€” is a proposed mechanism by which leaky gut contributes to autoimmune disease onset and progression.
๐Ÿ˜ด
Sleep Disruption
Gut bacteria produce GABA precursors and influence melatonin synthesis. A dysbiotic, inflamed gut disrupts both. Many cases of chronic insomnia have an unaddressed gut microbiome and permeability component at the root.
โšก
Chronic Fatigue
The immune system's constant inflammatory response to gut-leaked particles is metabolically exhausting. Many cases of unexplained chronic fatigue trace back to the energy burden of chronic, low-level gut-driven immune activation.

"BPC-157 wasn't designed for the gut โ€” it was isolated from it. The body had already solved this problem. We just learned how to use what it made."

The Peptide Stack

Peptides Used in Gut Health & Immunity

Five peptides covering the full gut-immune spectrum โ€” barrier repair, mucosal anti-inflammation, immune modulation, systemic repair, and the circadian foundation the gut microbiome depends on.

BPC-157
โ˜… The Gut Anchor
Discovered in gastric juice โ€” the only peptide that was literally born in the gut. Repairs tight junctions, promotes mucosal angiogenesis, normalizes gut motility, and supports the vagal tone that connects gut to brain. Genuinely oral bioavailable โ€” capsules are clinically meaningful for gut-targeted use.
Full Profile
KPV
Mucosal Anti-Inflammatory
Tripeptide fragment of ฮฑ-MSH with direct anti-inflammatory effects in the gut mucosa. Reduces TNF-ฮฑ and IL-6 at the mucosal level. Synergistic with BPC-157 โ€” BPC-157 repairs the barrier; KPV calms the immune response driving the damage. Essential for active IBD presentations.
Profile
Thymosin Alpha-1
Immune Modulation
Thymus-derived immune regulator. Raises underactive immune responses and calms overactive ones โ€” genuine bidirectional modulation. Directly relevant to the immune dysregulation underlying autoimmune conditions and Long COVID. Works alongside BPC-157 for the gut-immune protocol.
Profile
TB-4 Fragment
Systemic Repair
Oral Thymosin Beta-4 fragment. Works systemically where BPC-157 acts locally. Supports immune balance and drives cell migration throughout the body โ€” particularly relevant when gut dysfunction has created systemic consequences beyond the gut itself.
Profile
Epithalon
Circadian Foundation
The gut microbiome runs on its own circadian clock โ€” disrupted sleep disrupts microbial composition. Epithalon restores deep sleep and circadian regulation, providing the temporal foundation that gut microbiome health depends on. Near-universal inclusion in gut health protocols.

Where to Start

The Gut Health Protocol โ€” In Order

Foundations first โ€” peptides cannot repair a gut that's still being actively damaged. The sequencing matters as much as the compounds.

Step 0 โ€” Remove Triggers
No protocol holds if the damage source is still active. Reduce or eliminate NSAIDs (primary gut lining disruptor), alcohol, ultra-processed foods, and known food sensitivities during the healing phase. If symptoms persist despite dietary cleanup, test for SIBO โ€” bacterial overgrowth actively undermines gut barrier repair and must be addressed concurrently.
Step 1 โ€” Start BPC-157
Oral, on an empty stomach, 30 minutes before first meal. This maximizes mucosal contact time before food dilutes or moves the peptide along. 250โ€“500 mcg/day. Most people notice motility normalization (less bloating, more regular bowel movements) within days; structural tight junction repair builds over 4โ€“8 weeks. The fastest first signal that BPC-157 is working is often mood improvement โ€” the vagal tone and serotonin benefits come online quickly.
Step 2 โ€” Add KPV
For active mucosal inflammation โ€” IBD, active colitis, or significant inflammatory gut presentations. KPV 500 mcgโ€“1 mg/day alongside BPC-157. BPC-157 repairs the structural barrier; KPV simultaneously reduces the mucosal immune activation that's driving the damage. For straightforward leaky gut without active IBD, KPV is optional โ€” BPC-157 alone is typically sufficient.
Step 3 โ€” Immune Layer
Thymosin Alpha-1 for immune dysregulation. When labs show elevated TGF-ฮฒ1, abnormal immune markers, or when autoimmune conditions are part of the picture, Thymosin Alpha-1 provides the immune-modulating layer that BPC-157 + KPV alone don't address. Works synergistically โ€” gut barrier repair removes the immune trigger; Thymosin Alpha-1 rebalances the immune response that was dysregulated by it.
Step 4 โ€” Circadian Reset
Epithalon for the gut microbiome clock. A 10-day Epithalon course restores deep sleep and circadian signaling โ€” the temporal framework that gut microbial diversity depends on. Sleep disruption is simultaneously a consequence and a cause of gut dysbiosis; Epithalon breaks this cycle from the circadian end, often producing measurable improvements in both sleep quality and digestive regularity within the first week.
Step 5 โ€” Foundations Ongoing
Peptides build on foundations โ€” not instead of them. Continue the 3P protocol (prebiotics + probiotics + postbiotics), L-glutamine 4โ€“10g/day as enterocyte fuel, zinc carnosine for mucosal repair, and the anti-inflammatory diet throughout and after the peptide course. Gut healing is a months-long process; the peptides accelerate it, but the foundations sustain it.
